چکیده
Glucose 6-phosphate dehydrogenase from Zeuconostoc mesenteroides, previously isolated in crystalline form (Olive, C., and Levy, H. R., Biochemistry, 6, 730 (1967)), is shown to be essentially homogeneous by disc gel electrophoresis and sedimentation velocity analysis. The weight average molecular weight is 103,700, determined by both high speed and low speed sedimentation equilibrium techniques at pH 4.6, which is the approximate isoelectric point. The partial specific volume was determined to be 0.717 at this pH. The enzyme possesses an absorption spectrum showing a maximum at 280 5 run with E&iTb = 1.15 and considerable fine . structure in the region of 250 to 270 nm.

Modification of glucose-6-phosphate dehydrogenase from Leuconostoc mesenteroides with the 2',3'-dialdehyde derivative of NADP+ (oNADP+).
Glucose-6-phosphate dehydrogenase from Leuconostoc mesenteroides is irreversibly inactivated by the 2,3'-dialdehyde of NADP+ (oNADP+) in the absence of substrate. The inactivation is first order with respect to NADP+ concentration and follows saturation kinetics, indicating that the enzyme initially forms a reversible complex with the inhibitor followed by covalent modification (KI = 1.8 mM). متن کاملCoupling of Reduced Pyridine Nucleotide in Leuconostoc Mesenteroides.
The demonstration that oxidation-reduction reactions mediated by the pyridine nucleotide coenzymes occur with the transfer of hydrogen between substrate and coenzyme (1) has led to the use of hydrogen isotopes to measure the occurrence of the specific coupling of reactions in the intact cell and organism (2-4). Several problems associated with the cellular organization and integration of the fe...
متن کاملCloning of the gene and amino acid sequence for glucose 6-phosphate dehydrogenase from Leuconostoc mesenteroides.
Amino acid sequencing of glucose 6-phosphate dehydrogenase (Glc6PD) from Leuconostoc mesenteroides yielded sequence for over 75% of the protein. Two oligonucleotides based on the amino acid sequence were used to isolate a partial Glc6PD gene clone (pLmz delta N65), from a pUC9 library, containing 85% of the coding sequence and the 3'-untranslated DNA, but lacking the 5'-noncoding DNA sequence a...
متن کامل6-Phosphogluconate dehydrogenase from leuconostoc mesenteroides.
The pathways for degradation of 6-phosphogluconate have been rather clearly defined for several organisms, the most notable of which are yeast (Horecker, 1953), Escherichia coli (Cohen, 1951), Pseudomonas saccharophila (Entner and Doudoroff, 1952; MacGee and Doudoroff, 1954), and Pseudomonas fluorescens (Kovachevich and Wood, 1954). The enzymes from yeast and E. coli appear to be similar, if no...
متن کاملImprovement of thermal stability of Leuconostoc pseudomesenteroides glucose-6-phosphate dehydrogenase
Glucose-6-phosphate dehydrogenase (G6PDH, EC 1.1.1.49) from Leuconostoc pseudomesenteroides (the previously named: Leuconostoc mesenteroides) catalyzes the oxidation of glucose-6-phosphate in the presence of the coenzyme NADP or NAD. The enzyme is one of the industrially important enzymes and is applied in clinical settings. It is mainly useful in determinations of glucose and creatine kinase b...
